<p>PURPOSE:To enable mass-culture of a callus of Crocus genus plant and to obtain a substance such as safranal, quercetin, etc., useful as an emmenagogue, spice, etc., by culturing the callus in an MS medium added with phytohormones. CONSTITUTION:A sterilized tissue slice of a plant of Crocus genus such as saffron, Colchicum autumnale, crocus, etc., is implanted in a medium produced by adding a slight amount of phytohormones such as 2,4-D (0.3-3ppm), NAA (0.3-10ppm), cytokinin, abscisic acid, ethylenegibberellin, kinetin, zeatin, benzyladenine, 2-isopentenyladenine, etc., to an MS (Murashige-Skoog) medium or in a conventional agar medium. After the callus induction for about 1 month, the obtained callus is transplanted to an MS medium added with the above hormones and cultured for about 1 month to obtain a callus of about 10 times the volume of the starting callus.</p> |
