PURPOSE:To obtain the title probe with enhanced marker level leading to higher sensitivity, to be used for diagnosis of hereditary disease by annealing with template DNA a single-stranded RNA having a part complementary to a nucleic acid to be detected and another part complementary to the template DNA followed by forming the DNA complementary to said template DNA to effect labelling. CONSTITUTION:Single-stranded RNA with single-stranded part complementary to a nucleic acid to be detected and another part complementary to part of template DNA being in succession is annealed with the template DNA followed by forming DNA complementary to the template DNA succeeding the RNA annealed by making an action of DNA polymerase large fragment. Thence, a detectable marker is directly introduced into the DNA part thus formed, or a compound capable of binding to a detectable compound is introduced thereinto and this compound is bound to a marker, thus obtaining the objective nucleic acid probe. |
