摘要 A method of isolating a random peptide-scaffold peptide fusion protein that binds a predetermined target protein is disclosed. First, a library of nucleotide sequences encoding random amino acid sequences are generated and cloned into a first expression vector to make a library test vector. The nucleotide sequence is expressed within a yeast cell as a test fusion protein containing a transcriptional activation domain, a random peptide domain, and a scaffold peptide domain. The DNA sequence encoding a target protein cloned into a second expression vector provides a target vector. The expressed target fusion protein contains a DNA-binding domain and a target protein. Test fusion protein-target fusion protein interaction is detected according to a two-hybrid protein-protein interaction assay. Also disclosed are an HIV-1 Rev protein binding peptide isolated by the method of the invention and methods of using the Rev interacting protein.
