Also provided are methods to detect the presence or amount of two or more vitamin D metabolites in a single assay.

A method for determining the amount of a vitamin D metabolite in a sample by tandem mass spectrometry, comprising: (a) ionizing the vitamin D metabolite to generate a precursor ion; (b) generating one or more fragment ions from the precursor ion having a mass-to-charge ratio of 251.30±0.5, 211.35±0.5, 209.20±0.5, or 179.10±0.5; (c) detecting the amount of one or more of the ions generated in step (b) and relating the detected ions to the amount of the vitamin D metabolite in the sample.
