A CRYSTALLIZED RECOMBINANT HUMAN HSPOT AND THE METHOD FOR CRYSTALIZING THE SAME

A recombinant protein SpoT(hSpoT) which is an enzyme for hydrolysis of growth suppression gene (malnutrition signal transduction material) ppGpp (guanosine 5'-diphosphate 3'-diphosphate) is provided to used for researching tertiary structure of protein. A recombinant protein hSpoT from human genome is crystallized through hanging-drop vapor diffusion method. The recombinant protein hSpoT comprises an amino acid sequence of the sequence number 1(SEQ ID NO:1) and has six histidines at the amino terminal. The hanging drop vapor diffusion method comprises: a first step of mixing purified protein solution and reservoir solution in a ratio of 1:1 to form drop; a second step of sealing the drop on prepared plate; and a third step of placing mixture drop of reservoir solution and protein having lower concentration than the reservoir solution at 15-25‹C for one to ten days to form hSpoT protein crystal.
