PURPOSE:To obtain a cloning vector enabling the use of actinomycetes as the host, by imparting a tetracycline-resistant gene having pock-forming capability to a specific bacterial strain. CONSTITUTION:A pock-forming capability is imparted to Streptomyces griseus ATCC10137, and a DNA fragment containing a tetracycline-resistant gene originated from Streptomyces rimosus is introduced into one site sensitive to the restriction enzyme BamHI. The above DNA fragment is obtained by the partial digestion of the DNA of Streptomyces ATCC10970 with the restriction enzyme Sau3AI. The hybrid plasmid can be produced by mixing and bonding (A) the plasmid pOA15 (prepared by the alkali extraction of Streptomyces sp.OA15, FERM-P No.6889) obtained by the scission with BamHI with (B) the DNA of Streptomyces rimosus obtained by the partial digestion with Sau3AI to obtain DNA fragments having the molecular weight of 2-25kb, and tranforming Streptomyces griseus ATCC1037 with the product. |
