PURPOSE:To obtain human lysozyme, by culturing cell transformed with DNA having a synthetic gene for the expression of human lysozyme. CONSTITUTION:Although the DNA sequence of human lysozyme gene is not clear at present, the gene of human lysozyme was designed and chemically synthesized based on the human mammary lysozyme. In the above process, the optimum sequense was determined according to the criteria that (i) a codon most permissible in a yeast supposed to be suitable for the expression of synthetic gene is vsed, (ii) a specific site recognized by a restriction enzyme (Xbal) is inserted in the gene to make an index in cloning or to facilitate the genetic modification after the construction of expression vector, and (iii) the self- complementary sequence or sequence complementary to other sequence on either chain or on double-chain is avoided as far as possible. |
