NEW MATERIAL:Lignin peroxidase L-II having following physicochemical properties. Action: generates veratryl aldehyde by oxidation of veratryl alcohol in the presence of hydrogen peroxide, and oxidatively decomposes diallylpropane in the presence of hydrogen peroxide; substrate specificity: oxidatively decomposes dimer model compound of lignin such as diallylpropane and veratryl glycerol-beta-guaiacyl ether, or oxidizes veratryl alcohol; optimum pH: 3; stable pH: 4 to 6; optimum temperature: 30 deg.C; heat stability: stable below 45 deg.C; isoelectric point: 4.0; molecular weight of the objective enzyme: 41000 (SDS polyacrylamide gel electrophoresis); the objective enzyme is strongly inhibited by EDTA or KCN, etc. USE:Enzyme making lignin to low molecular weight. PREPARATION:Microorganism belonging to genus Bjerkandera such as Bjerkandera adusta IFO 5307 is inoculated in medium. |
