The present invention relates to a carboxy-terminus truncated beta-galactosidase enzyme variant having high transglycosylation activity, and a use thereof and more specifically, to: a carboxy-terminus truncated beta-galactosidase variant; a gene coding for the same; a recombinant vector containing the gene; a recombinant microorganism, which has been genetically transformed by the gene or the recombinant vector; and a method for preparing a beta-galactosidase variant by using the recombinant microorganism. By using high transglycosylation activity of the carboxy-terminus truncated beta-galactosidase variant according to the present invention, galactooligosaccharide can be efficiently mass-produced.
