摘要 PURPOSE: To conduct the subject identification and detection of bacteria useful for diagnosing infectious diseases, development of the microbial environmental cleanup system and food production process control, etc., quickly in high accuracy by using the nucleic acid base sequence of a RNA polymerase σ-factor gene as indicator. CONSTITUTION: Using the nucleic acid base sequence of a RNA polymerase σ-factor gene markedly varied even among the same families and strains of a kind of bacteria as indicator, a DNA fragment containing a specific common base sequence among RNA polymerase σ-factor genes of bacteria is amplified by PCR process using a sense primer containing a base sequence coding an amino acid sequence expressed by the formula MYMREMGTV as a part of the amino acid sequence on σ70 of Escherichia coil K12 strain and an anti-sense primer containing a base sequence coding an amino acid sequence expressed by the formula KKEMVEAN; subsequently, the base sequence of the DNA fragment is determined by e.g. dideoxy method, and based on this base sequence, the objective identification and detection of the bacteria is easily and quickly accomplished in high accuracy.
