Polymer-coupled peptidases

Detection or quantitative estimation hemostasis peptidase inhibitor in a sample comprises contacting the inhibitor in the sample with a polymer-coupled hemostasis peptidase, and measuring the activity of polymer-coupled peptidase after contacting with the inhibitor in the sample and comparing with one or more reference values to determine the inhibiting activity or measuring the clotting time of the sample after contacting with the coupled peptidase and comparing the clotting time with one or more reference values to determine the change in the clotting time. Independent claims are included for: (1) an in vitro method for neutralization of the inhibitory activity of the hemostasis peptidase inhibitor comprising contacting a sample containing the inhibitor with a polymer-coupled hemostasis peptidase; (2) polymer-coupled hemostasis peptidase for use in the restoration of clotting ability of the blood, for increasing the clotting ability of the blood and/or for increasing the blood clotting; (3) a method for modifying the specificity of hemostasis peptidase so that the peptidase loses its reactivity in hemostasis comprising coupling the peptidase to a polymer; (4) a device, for separating the hemostasis peptidase inhibitor from a sample or blood circulation of a patient, comprising a polymer-coupled hemostasis peptidase; and (5) a test kit comprising the polymer-coupled hemostasis peptidase as first reagent and a substrate for peptidase as second reagent.
