TRANSGENIC ANIMAL WITH DEFECTIVE PROTEOLIPID PROTEIN AND MANUFACTURE OF SAID ANIMAL

摘要

PURPOSE: To obtain a transgenic nonhuman mammal lacking a myelination process which is useful as an animal model of multiple sclerosis by introducing antisense RNA by minute mutation. CONSTITUTION: An intrinsic proteolipid protein (plp; 30 kD) gene on a X- chromosome is exchanged by homologous recombination in embryo stem cells with specific pip and isoproteolipid protein (DM 20; 26 kD) structure having neomycin-resistant gene with antisense direction by intron III. By the presence of antisense RNA, the obtd. transgenic nonhuman mammal such as a mouse can not produce a proteolipid protein to compact the myelin in the central nervous system.
