A method of concentrating nucleic acid molecules in a urine sample from a patient, wherein the nucleic acid molecules have a size the same or greater than a target nucleic acid molecule, the method comprising a) obtaining the sample of urine from the patient, wherein the initial sample has an initial volume of about 20_ml or greater; b) applying the sample to a membrane having a molecular weight cut-off that is about 20% to 50% smaller than a target nucleic acid molecule; c) applying a pressure of about 0 to about 5 bar, or about 0 to about 70_psi to the sample in contact with the membrane; and d) collecting a retentate, wherein the retentate comprises a 5-fold or greater concentration of a target nucleic acid as compared to its initial concentration in the urine.
