The disclosure relates a method of making a piperazine derivative compound of formula (IV), wherein the variable R is as defined in the specification. The disclosure also relates to a composition comprising: a sterol, a neutral lipid, a PEG or PEG-modified lipid, and a lipid which contains a piperazine moietie of such compounds, and optionally a targeting lipid comprising N-acetyl galactosamine as a targeting moiety. These lipid compositions are useful for therapeutic agent delivery of nucleic acids as well as nucleic acid-lipid particle compositions. |
