摘要 The invention relates to recombinant microorganisms that have been engineered to produce various chemicals using genes that have been repurposed to create a reverse beta oxidation pathway. Generally speaking, the beta oxidation cycle is expressed and driven in reverse by modifying various regulation points for as many cycles as needed, and then the CoA thioester intermediates are converted to useful products by the action of termination enzymes.

主权项 1) An engineered microorganism, comprising: a) overexpression of β-oxidation cycle enzymes as compared to a wild type, said enzymes comprising: i) a thiolase catalyzing the conversion of (Cn)-acyl CoA to β-ketoacyl-CoA;ii) a hydroxyacyl-coA dehydrogenase catalyzing the conversion of β-ketoacyl-CoA to β-hydroxyacyl-CoA;iii) an enoyl-coA hydratase catalyzing the conversion of β-hydroxyacyl-CoA to trans-Δ2-enoyl-coA; andiv) an acyl-CoA dehydrogenase or a transenoyl-CoA reductase catalyzing the conversion of trans-Δ2-enoyl-coA to (Cn+2)-acyl CoA; b) functional operation of a β-oxidation cycle in a reverse/biosynthetic direction, as recited in enzyme reactions i) through to iv); and c) overexpression of one or more termination enzyme(s) as compared to a wild type, wherein one or more termination enzyme(s) converts a reverse beta oxidation cycle intermediate to a product selected from the group consisting of a trans Δ2 fatty alcohol, a β-keto alcohol, a 1-3 diol, a β-hydroxy acid, a carboxylic acid, an alkane and an alkene.
