The present invention provides novel compositions and methods for creating quantitative standards to calibrate analytes. These compositions and methods enable the creation of standards and calibrators for analyzing analytes and measuring clinical biomarkers. Also provided are kits comprising the novel compositions for use in assays, for example sandwich immunoassays.

An isolated, modified peptide comprising; an N-terminal immunoreactive domain of Aβ42, a central aggregation domain replacing amino acids 17-20 and adjacent external peptides by a linker that is a non-immunoactive domain peptide selected from the group consisting of polyethylene glycol, a lipid, a globular protein, a nucleic acid and an alkyl chain, and a C-terminal immunoactive domain of Aβ42; wherein said peptide is a non-aggregating peptide.
