A total internal reflection fluorescence imaging system (100) and a sequencing device, the total internal reflection fluorescence imaging system (100) comprising a first imaging system (10) and a second imaging system (20), the first imaging system (10) comprising N separate laser optical paths, the second imaging system (20) determining, by means of the difference in position at which a second receiving device (24) acquires a second image, a change in the height of a sample, and performing automatic compensation according to the change in the height of the sample, so as to ensure that the sample is always at the focus position. By means of the second imaging system (20) respectively performing optical path adjustment of the N separate laser optical paths, the N separate laser optical paths are enabled to have the same penetration depth, and therefore no further optical path adjustment is required when a different light source is used, thereby economizing on time-consuming optical path adjustment during experiments. |
