Preparation and use of a polymeric microfludic device for nucleic acid purification. The microfluidic device comprises a randomly roughened polymeric surface fabricated with gaseous plasma etching. The design of the microfluidic chip encompasses on chip mixing of the samples inside the microfluidic, which together with the roughness render the microfluidic device suitable for Lab-on-a-chip (LOC) systems for on-chip nucleic acid extraction and purification.
