A method of generating a pseudotyped lentiviral vector particle comprising: (a) culturing in a culture medium comprising kifunensine a virus packaging cell comprising: (1) a lentiviral vector genome comprising an exogenous polynucleotide of interest, (2) a polynucleotide encoding a Sindbis E2 glycoprotein that preferentially binds dendritic cells expressing DCSIGN, and (3) a polynucleotide encoding a Vpx protein or a Vpr protein that retains SAMHD1-inhibiting activity; and (b) isolating a pseudotyped lentiviral vector particle that preferentially binds dendritic cells expressing DC-SIGN, and compositions thereof. |
