PROBLEM TO BE SOLVED: To provide: a reagent capable of dissolving acid-fast bacteria contained in a sample to extract nucleic acid (particularly RNA) with a simple way at a short time period without requirement of a specific device or a troublesome operation; and a method of detecting the acid-fast bacteria contained in the sample with the reagent.SOLUTION: The above mentioned problem is solved with: a reagent for dissolving acid-fast bacteria and extracting its RNA, consisting of a solution comprising alkylbenzene sulfonic acid and a neutralization agent which produces no metal salt of the acid in a reaction with the acid; a method of extracting RNA in which the reagent is added to a sample comprising the acid-fast bacteria to perform a heating process and to dissolve the acid-fast bacteria; and a method of detecting the acid-fast bacteria in the sample by amplifying and detecting RNA derived from the acid-fast bacteria obtained with the method with a primer/probe specific to the RNA. |
