Methods of modulating high mannose glycoform content of a protein in a cell culture by contacting the cells expressing the protein of interest with monesin are provided.

establishing a mammalian cell culture expressing the recombinant protein in a serum-free culture medium in a bioreactor, maintaining the mammalian cells during a production phase, and contacting the cell culture with monensin.
