摘要 An objective of the present invention is to provide a glycan releasing method which can be applied to construction of a system for automation of glycan analysis, and particularly a glycan releasing method capable of analyzing an O-linked glycan. The objective could be achieved as a result of finding that the pH is lowered by using an ammonium salt or ammonium ion in the absence of concentrated aqueous ammonia, not using concentrated aqueous ammonia, thus drastically avoiding an undesired side reaction such as a peeling reaction or the like. Therefore, the present invention provides a method of producing an O-linked glycan from a glycan-binding substance having the O-linked glycan, the method includes the steps of (A) bringing an ammonium salt or ammonium ion into contact with the glycan-binding substance in the absence of concentrated aqueous ammonia (for example, under the conditions of the pH of about 7 or higher and lower than about 11); (B) neutralizing the reaction solution obtained in step (A); and (C) collecting the released glycan.
