The purpose of the present invention is to establish an extraction method for preparing a Halocynthia roretzi extract with improved anti-inflammatory effects and immunomodulatory activities by solving problems including human toxicity, low extraction yield, high expenses, and others as problems of conventional extraction methods, and by increasing contents of polysaccharides in the extract. The present invention relates to a method for preparing a Halocynthia roretzi extract with improved immunomodulatory activities comprising a step of adding water and a protein hydrolysis enzyme to a pretreated Halocynthia roretzi shell, extracting the mixture to obtain an extract, and separating the extract using an ion-exchange chromatography; to a Halocynthia roretzi extract with improved immunomodulatory activities prepared by the method; and to a processed food containing the Halocynthia roretzi extract.
