The present invention relates to a method for promoting directed differentiation of human pluripotent stem cells into retinal pigment epithelium (RPE) fate, the method comprising: a. culturing human pluripotent stem cells in a culture system comprising a basic medium supplemented with nicotinamide (NA) to generate differentiating cells; and b. culturing said differentiating cells in a culture system comprising a basic medium supplemented with one or more members of transforming growth factor ² (TGF ²) superfamily, whereby said human pluripotent stem cells are induced to differentiate into RPE fate.
