Genetically modified cells, tissues, and organs for treating or preventing diseases are disclosed. Also disclosed are methods of making the genetically modified cells and non-human animals.

1. A genetically modified animal with reduced protein expression of one or more first genes, wherein said genetically modified animal is a member of the Laurasiatheria superorder or is a non-human primate, and wherein said one or more first genes comprise:
a) a component of a major histocompatibility complex (MHC) I-specific enhanceosome, b) a transporter of an WIC I-binding peptide, or c) complement component 3 (C3), wherein said reduced protein expression is in comparison to a non-genetically modified counterpart animal.
