PROBLEM TO BE SOLVED: To provide a nucleic acid amplification reaction device and a nucleic acid amplification method in which sufficient nucleic acid amplification reaction products can be obtained even in the case where reaction time is short.SOLUTION: A nucleic acid amplification reaction device includes: an installing part which is capable of installing a nucleic acid amplification reaction container filled with nucleic acid amplification reaction liquid and liquid which has smaller specific gravity than the nucleic acid amplification reaction liquid and does not mix with the nucleic acid amplification reaction liquid; a first heating part which heats a first region of the nucleic acid amplification reaction container to a first temperature; a second heating part which heats a second region of the nucleic acid amplification reaction container to a second temperature; and a drive mechanism which switches a first arrangement in which the first region comes under the second region in a working direction of gravity and a second arrangement in which the second region comes under the first region in a working direction of gravity. The magnesium ion concentration of the nucleic acid amplification reaction liquid is 1.8 to 9.0 mM.SELECTED DRAWING: None
