PROBLEM TO BE SOLVED: To provide potent anti-EphA3 antibodies, and to provide methods of using the antibodies for the treatment of a disease involving EphA3.SOLUTION: This invention relates to: high-affinity antibodies to EphA3 that have reduced immunogenicity when administered to a human to treat diseases; and a method of using the antibodies. The antibody comprises a VH region that comprises a CDR3 comprising an amino acid sequence X1GX2YEX3FDX4, wherein X1 is S or G, X2 is Y or V, X3 is E or D, and X4 is S, V, or I, with the proviso that: when the amino acid sequence is SGYYEDFDS, the CDR1 is not SYWIN; and when the amino acid sequence is SGYYEEFDS, the CDR1 is not TYWIS. The antibody has: a J segment having no more than two amino acid changes relative to a human germline J segment amino acid sequence; and at least 90% identity to a V segment amino acid sequence.SELECTED DRAWING: Figure 2 |
