A substrate and manufacturing method thereof, and display device, the substrate comprising: a substrate (101); multiple gate lines (103) disposed in parallel; a first insulation layer (102) covering the gate lines (103); multiple data lines (104) perpendicular to the gate lines (103) and located at the first insulation layer (102); a second insulation layer (105) covering the data lines (104); a pixel electrode (106) of a sub-pixel area surrounded by the data lines (104) and the gate lines (103); a light deflecting film (107) covering the pixel electrode (106); and an auxiliary gate line (108) parallel to the gate lines (103) and disposed at the second insulation layer (105); at least two locations on each first auxiliary gate line (108) are electrically connected by sequentially passing through apertures (109) of the first insulation layer (102) and the second insulation layer (105) to two corresponding locations on the gate lines (103); the first auxiliary gate line (108) and the light deflecting film (107) are formed via the same patterning process in the same layer of a transparent conductive material. The substrate can reduce a signal delay in a display device and is used for manufacturing a display.
