FIELD: chemistry.SUBSTANCE: method of extracting placental alkaline phosphatase by homogenising placental tissue to a homogeneous mass, extracting tris-HCl with a butanol-containing buffer, centrifuging said mixture after a day with diethyl ether, collecting the aqueous phase and then performing affinity chromatography of the placental alkaline phosphatase on immobilised fresh-water sponge lectin, followed by washing with NaCl solution buffered with tris-HCl buffer, and eluting the placental alkaline phosphatase bound on the column with lactose solution in a borate buffer under certain conditions.EFFECT: method simplifies and speeds up extraction of placental alkaline phosphatase, increases the output and the degree of purity thereof.2 dwg, 2 tbl, 2 ex |
