PROBLEM TO BE SOLVED: To provide a method for identifying/isolating polypeptides with desirable biophysical properties, such as solubility, stability, high expression, monomericity, binding specificity or non-aggregation, using a high throughput method for screening polypeptides.SOLUTION: The invention provides a method useful as an immunotherapy or a diagnostic agent comprising obtaining a phage display library; allowing infection of a bacterial lawn by the library phage; identifying phage which forms larger plaque than the average plaque on the bacterial lawn; identifying sequences of monomeric human Vand human V, and multimeric complexes of human Vand human V; making additional library for identifying additional polypeptide using identified Vand V; and further subjecting Vand Vto DNA shuffling to select for improving biophysical properties.SELECTED DRAWING: Figure 1 |
