PROBLEM TO BE SOLVED: To provide a method, which is more efficient than the conventional method, for culturing human pluripotent stem cells while maintaining undifferentiated condition; and to provide a kit therefor.SOLUTION: The method of the invention comprises (a) culturing human pluripotent stem cells in a first culture medium for human pluripotent stem cells containing activin; (b) replacing the first culture medium with a second culture medium for human pluripotent stem cells containing no activin to culture the human pluripotent stem cells; and (c) passaging the human pluripotent stem cells in the first culture medium. We confirmed that pluripotency of the stem cells is maintained at a high rate irrespective of the skill level of an experimenter's cultural technique when subsequently the above (b) and (c) are repeated successively.SELECTED DRAWING: None |
