Methods for affinity maturation-based antibody optimization

Provided herein is a rational method of affinity maturation to evolve the activity of an antibody or portion thereof based on the structure/affinity or activity relationship of an 5 antibody. The resulting affinity matured antibodies exhibit improved or optimized binding affinity for a target antigen.
