The present invention provides a method for modifying a targeted site of double-stranded DNA in a cell, wherein the method includes a step in which: a complex, obtained by bonding a nucleic acid sequence recognition module for bonding specifically with a target nucleotide sequence in selected double-stranded DNA and DNA glycosylase having sufficiently low reactivity with DNA that has an undistorted double helix structure (unrelaxed DNA), is brought into contact with the double-stranded DNA; and one or more nucleotides of the targeted site are converted into one or more other nucleotides or are deleted, or one or more nucleotides are inserted at the targeted site, without cleaving at least one of the strands of the double-stranded DNA at the targeted site. |
