The present invention relates to an agent for prevention or treatment of diabetes using glucagon regulation based on an olfactory receptor expressed in the pancreas and an olfactory receptor of the pancreatic islet. According to the present invention, an olfactory receptor is identified to be expressed in an α-cell of the pancreas, and it was identified that by a ligand of the olfactory receptor, intracellular Ca2+ mobilization in the pancreas is regulated and finally glucagon secretion of the pancreas can be regulated, and glucagon secretion decreased when the olfactory receptor of the pancreas was treated in a high glucose condition such as actual diabetes.
