A method for creating initial breeding forms of a fusariose-resistant cucumber, including screening of genotypes in the culture of isolated tissues in vitro on MS nutrient medium, modified as a selective background by 40% mixture of filtrates of cultural liquid (FCL) of two species of fungi (F. oxysporum f. sp. cucumerinum and F. solani) at a ratio of 2:1, which corresponds to a natural involvement in the pathogenesis of this disease, the selection of genotypes that exceeded reference samples with high level of resistance to the disease by morphometric parameters thereof. The apical meristems from sterile seedlings are planted on the induction nutrient medium adding a selective agent thereto. Selection of tracheomycosis-resistant genotypes in culture in vitro is carried out directly on the differentiated plant organism after 28 days of cultivation on selective medium at lighting conditions of 2000 lux and at a temperature of 23-25 °C. Assessment of the resistance of plant-regenerants to the effect of FCL in culture in vitro is carried out by determining the resistance index (RI) expressed as a percentage. For further selective use as a source of resistance the samples that exceeded by the index of resistance the reference genotypes with determined high field resistance to Fusarium wilt are propagated. |
