Antibodies recognizing a carbohydrate containing epitope on CD-43 and CEA expressed on cancer cells and methods using same

The present invention provides antibodies (such as chimeric and humanized antibodies) specifically bind to an epitope on CD43 and CEA expressed on nonhematopoietic cancer cells. In addition, the present invention also provides use of the antibodies described herein for diagnostic and therapeutic purposes.

1. A polynucleotide comprising a nucleic acid sequence encoding an antibody comprising a heavy chain and a light chain, wherein
(a) the heavy chain comprises a heavy chain variable region comprising three complementary determining regions (CDRs) from the amino acid sequence of SEQ ID NO:1 and a heavy chain constant region comprising the amino acid sequence of SEQ ID NO:9, wherein the hinge region of the heavy chain constant region comprises at least one amino acid insertion, deletion or substitution; and (b) the light chain comprises a light chain variable region comprising three complementary determining regions from the amino acid sequence of SEQ ID NO:2 and a light chain constant region comprising the amino acid sequence of SEQ ID NO:10 or a light chain constant region comprising the amino acid sequence of SEQ ID NO:10 and further comprising at least one amino acid insertion, wherein the antibody specifically binds to a carbohydrate epitope on a CD43 or a carcinoembryonic antigen (CEA) expressed by the nonhematopoietic cancer cells.
