The present invention relates to compounds and methods for imaging pancreatic beta-cells and determination of pancreatic beta-cell mass (BCM). The compounds comprises a peptide having an insulin binding amino acid sequences, a cell peptide having a penetrating amino acid sequence, and a ligand, e.g. a radioligand or a fluorescent ligand. The methods provides live cell staining for pancreatic islets and beta cells in vitro or in vivo for diagnosis and monitoring the efficacy of treatments for pancreatic beta cell disorders, e.g. Type 1 and Type 2 diabetes.
