The invention relates to the microbiological industry, microbiology and biotechnology, and specifically to the production of physiologically active compounds, and concerns the production of a protein, bacteriorhodopsin, by means of microbiological synthesis. A method for producing bacteriorhodopsin involves cultivation in a fermenter with a volume of 10 litres over a period of 6 days, and separation and breaking down of a biomass during incubation in distilled water over a period of one day with constant stirring. The separation of the precipitate is carried out by centrifugation at 10000 g for 15 minutes; the obtained supernatant is centrifuged at 50000 g for 55 minutes; the supernatant is discarded; and distilled water is added to the obtained precipitate in the same volume; the precipitate is suspended in water 3-4 times, whereupon it is applied to a chromatography column with a silica gel in a ratio of 200 absorbance units in the applied sample, at a wavelength of 280 nm, to 100 ml of silica gel. After elution, samples are taken, with a ratio of the absorbance of the solution at a wavelength of 280 nm to the absorbance of the solution at a wavelength of 570 nm (D280/D570) of less than 2.5. When the strain Halobacterium salinarum VKPM B-11953 is used, a high yield of bacteriorhodopsin is achieved. |
