A method for obtaining a mutant oleaginous yeast strain which comprises endogenous acyl-CoA:diacylglycerol acyl transferases, an endogenous acyl-CoA oxidase, an endogenous oleate desaturase, an endogenous multifunctional enzyme having a 3-hydroxyacyl-CoA dehydrogenase and enoyl-CoA hydratase activities, an endogenous 3-oxoacyl-CoA thiolase, an endogenous 2-methylcitrate dehydratase, and an endogenous phospholipid:diacylglycerol acyl transferase, wherein fatty acid beta-oxidation occurs within the yeast strain, and wherein the mutant oleaginous yeast strain is a template yeast strain for obtaining other mutant oleaginous yeast strains, said method comprising the steps of: (a) inhibiting the fatty acid beta-oxidation in said strain, (b) inhibiting the expression or of the activity of one or more of the endogenous acyl-CoA:diacylglycerol acyl transferases of said strain, (c) inhibiting the expression or the activity of the endogenous oleate desaturase—of said strain, and optionally (d) inhibiting the expression or the activity of the endogenous phospholipid:diacylglycerol acyl transferase of said strain.
