The present invention relates to a method for increasing mannosylphosphorylation using overexpression of YlMpo1, and more specifically, to a method for producing bi-mannosylphosphorylated sugar chain-added proteins by over-expressing YlMpo1 proteins in recombinant yeasts with no yeast-specific sugar chain biosynthetic pathway. The bi-mannosylphosphorylated sugar chains produced by the method of the present invention can be easily converted to two mannose-6-phosphate-added bi-phosphorylated sugar chains by an enzymatic or chemical method and then converted to sugar chains (P_2-Man_6-GlcNAc_2) which have a structure to provide a high bond affinity to mannose-6-phosphate receptors and excellent lysosome targeting capability by trimming using treatment with mannosidase. Accordingly, the present invention produces a high content of bi-mannosylphosphorylated sugar chain-added glucoproteins by over-expressing YlMpo1 in yeasts with no yeast-specific sugar chain biosynthetic pathway, thereby providing glucoproteins with excellent lysosome targeting capability. |
