Conversion of somatic cells to induced reprogrammed neural stem cells (irNSCs)

This application relates to a method for converting somatic cells to Neural Stem Cells (NSCs). Moreover this application relates to a method for converting human fibroblasts, keratinocytes or adipocytes to neural stem cells based on linked steps of genes transduction and chemically defined medium induction.

1. A method of producing Neural Stem Cells (NSCs), comprising:
a) providing human somatic cells selected from the group consisting of fibroblasts, adipocytes, or keratinocytes, b) reprogramming said cells to NSCs by introducing a first and a second gene, wherein the first gene is Sox2, and the second gene is selected from the group consisting of Bmi1, Mash 1, Sox11, Emx2, Foxg1 and Pax6; and c) culturing the cells of step (b) in medium comprising growth factors selected from the group consisting of FGF2, EGF and BDNF and a small molecule inhibitor of Rho-associated coiled-coil forming protein serine/threonine kinase (ROCK inhibitor), to produce NSCs.
