An array substrate and manufacturing method thereof, and display device; the manufacturing method of the array substrate comprises forming a conducting layer (20) electrically connecting two neighboring pixel electrodes (104) when forming the pixel electrodes (104), and particularly comprises: forming a photoresist layer (30) on a surface of a substrate formed with the conducting layer (20); forming a first photoresist removal area (A) and a photoresist remaining area (B) via a single exposure developing process, the first photoresist removal area (A) corresponding to the position of the conducting layer (20); etching the conducting layer (20) of the first photoresist removal area (A), forming an isolation area (106) for avoiding an electrical connection between the pixel electrodes (104) of two neighboring pixel units, and forming, in the isolation area (106), a metal wire having no electrical connection with the two neighboring pixel electrodes (104). The array substrate and manufacturing method thereof can avoid damage on too much thin film layers on the array substrate in the process of reducing bright dot defects.
