PROBLEM TO BE SOLVED: To identify an endoglucanase gene or a &bgr;-glucosidase gene by: isolating genomic DNA containing a cellulase gene that is classified as the endoglucanase or the &bgr;-glucosidase, from Acremonium cellulolyticus; and analyzing its base sequence.SOLUTION: Amino acid sequences of known endoglucanases and &bgr;-glucosidases are compared with each other assiduously, and an amino acid sequence which can be conserved in Acremonium cellulolyticus is found. Based on the information, various primers are designed. Using the various primers thus designed, a PCR is carried out employing genomic DNA or cDNA as a template. As a result, fragments of genes for endoglucanases and genes for &bgr;-glucosidases can be obtained. Thus primers are designed based on the gene fragments, the PCR is continued to amplify nine endoglucanase and &bgr;-glucosidase genes, and base sequences are analyzed to achieve the present invention.SELECTED DRAWING: None |
