The invention relates to the microbiology industry, to microbiology and biotechnology, and specifically to the production of physiologically-active compounds, and relates to the production of a protein, bacteriorhodopsin, by means of microbiological synthesis. A method for producing bacteriorhodopsin has been developed which includes cultivation in a 1200-liter fermenter for 7 days, and separation and destruction of biomass in a separator-module. Precipitate is separated via centrifugation at 10000 g for 15 minutes, the resulting supernatant is centrifuged at 50000 g for 55 minutes and discarded, and distilled water of the same volume is added to the produced precipitate; the precipitate is suspended in water 3-4 times, and is then applied to a chromatographic column such that 200 optical units are present in the applied sample, at a wavelength of 280 nm, per 100 ml of silica gel. Following elution, samples are selected which have a ratio of solution optical density at a wavelenth of 280 nm to solution optical density at a wavelength of 570 nm (D280/D570) of less than 2.5. The use of Halobacterium salinarum strain RNCIM B-1185 0 allows for producing a high yield of bacteriorhodopsin. |
