The present invention relates to a composition for alleviating aging or extending lifespan, comprising a Humulus japonicus leaf extract as an active ingredient. More specifically, a Humulus japonicus ethanol extract suppresses the generation of reactive oxygen species generated in skin cells by an inducer generating the reactive oxygen species and UVB, increases the expression level of SIRT1 that is a protein related to the lifespan extension, and extends lifespan of yeast and inhibits skin aging by activating AMPK. Therefore, the composition comprising the Humulus japonicus leaf extract as an active ingredient of the present invention can be used as a pharmaceutical composition for effectively alleviating aging or extending lifespan, a health food composition, or a cosmetic composition.
