The present invention relates to a preparation method of biodisel, which comprises: a first step for adding Fe^3+ ions to a microalgae culture medium and concentrating the microalgae; a second step for adding hydrogen peroxide to the microalgae concentration concentrated from the first step and extracting microalgae lipid; a third step for adding methanol to a microalgae lipid extraction extracted from the second step and inducing a first esterification reaction; and a forth step for adding an alkaline catalyst to the reaction solution which completed the first esterification reaction from the third step and inducing a second esterification reaction. The preparation method of biodisel according to the present invention is economical in aspects of costs and energy consumption and can manufacture biodisel in a short period of time at low costs.
