The present invention relates to a pharmaceutical composition comprising DUSP5 as an active ingredient for preventing or treating metabolic bone diseases, to a pharmaceutical composition for preventing or treating STAT3-mediated diseases, and to a method for inhibiting STAT3 activities by using DUSP5. DUSP5 of the present invention is capable of effectively inhibiting the activities or the expression of RANKL, RANK, NFATc1, and TRAP which are related to osteoclast differentiation, inhibiting or reducing Th17 activities, and promoting or increasing regulatory T cells (Treg) at the same time. In addition, DUSP5 can be usefully used as a novel drug, which can treat metabolic bone diseases and STAT3-mediated diseases, by inhibiting the STAT3 activities through the inhibition of the phosphorylation of STAT3.
