The present invention relates to the detection of Salmonella Typhi infection in a sample. In particular, the invention provides highly sensitive and specific primer pair useful for the detection of Salmonella Typhi. The invention also provides a PCR reaction mixture, detection kit and a method based on the same for the detection of Salmonella Typhi. The invention also provides a probe sequence useful for the detection of Salmonella Typhi infection. In one other important aspect of the invention, there is provided RNA aptamer and RNA probe corresponding to the primer sequences and DNA probe sequence respectively. The detection is performed from human blood samples in vitro.
