Quantitative determination of protein tyrosine nitration levels is used to determine the severity and progressive nature of a disease associated with nitration stress. Independent claims are also included for: (1) antibodies specific for albumin with nitrated tyrosine at position 138 or 411; (2) determining the severity and progressive nature of a disease associated with nitration stress by quantitatively determining the nitration level of a protein P1 by comparing the concentration of nitrated P1 with the total concentration of nitrated and nonnitrated P1 and comparing this nitration level with the nitration level of P1 in a sample from an individual that is not disease-affected; (3) preparing a protein with the same nitrated tyrosine residues as a nitrated protein occurring in a disease associated with nitration stress by reacting a protein with tetranitromethane in a tetranitromethane:tyrosine molar ratio of up to 20:1 in an aqueous buffer with a pH of 7.5-8.5 and identifying the nitrated tyrosines, especially by mass spectrometry. |
