Embodiments of the invention provide a method of genotyping a C. gattii sample, which can include forming a plurality of mixtures for nucleic amplification. The method can include amplification of specific sequences within the C. gattii genome that can provide definitive genotype information to distinguish between one or more types or subtypes of C. gattii.

1. A method of genotyping a Cryptococcus gattii sample, the method comprising the steps of:
adding a first oligonucleotide to a first mixture comprising the sample, wherein the first oligonucleotide added to the first mixture includes a sequence selected from the group consisting of SEQ ID NO. 7, SEQ ID NO. 8, and SEQ ID NO. 9; adding a first oligonucleotide to a second mixture comprising the sample, wherein the first oligonucleotide added to the second mixture includes a sequence selected from the group consisting of SEQ ID NO. 13, SEQ ID NO. 14, and SEQ ID NO. 15; subjecting the first and second mixtures to conditions that allow nucleic acid amplification; and determining the genotype of the Cryptococcus gattii sample on the basis of results of the nucleic acid amplifications of the first and second mixtures.
